Inimmunoelectrophoresisagainstpooledserumandenrichedserumproteinsfractionsprecipitationcanbeobservedofnotlessthan20individualproteinscomponents.InprecipitatingtechniquesasimmunoelectrophoresisandrADIalimmunodiffusion(Ouchterlony)toidentifytheserumproteinpattern,orthepresenceorabsenceofanindividualcomponent.Toevaluatethepurityofanisolatedserumproteinincludingimmunoglobulins.Sinceimmunoprecipitationdependsonacorrectantigen/antibodyconcentrationratio(zoneofequivalence)inthegelmedium,theproteinanalysisbyimmunoelectrophoresisofserumoranyotherBIOLOGicalfluidorproteinfractionshouldincludedifferentproportionsofthereactants.ItisnotpossIBLetoobtainanoptimalproteinpatterninasingleanalysis.Theelectroendosmosiseffectofdifferenttypesofagaronproteinswithadifferentnetchargecanbeusedtooptimizetheresolutionpowerofthetestsystem.AgarNordicNr.2containssufficientpositivelychargedionstooptimizetheresolutionoftheproteinsinthebeta-gammaregions,whilethealpharegionswillbecomemoredense.Highlypurifiedagar(AgarNordicnr.1)withlowelectroendosmosisfavourstheresolutionoftheproteinsinthealpharegions,whilethemajorcomponentsinthebeta-gammaregioncanstillbeidentified.

Inter-speciescross-reactivityisanormalfeatureofantibodiestoanimalproteinssincehomologousproteinsofdifferentspeciesfrequentlyshareantigenicdeterminants.ThisantiSerumhasnotbeenadsorbedforsuchcross-reactivity.Consequentlyitisnotspecies-specific.

Storage:

Thelyophilizedantiserumisshippedatambienttemperatureandmaybestoredat+4°C;prolongedstorageatorbelow-20°C.Dilutionsmaybepreparedbyaddingphosphatebufferedsaline(PBS,pH7.2).Repeatedthawingandfreezingshouldbeavoided.Ifaslightprecipitationoccursuponstorage,thisshouldberemovedbycentrifugation.Itwillnotaffecttheperformanceoftheantiserum.Dilutedantiserumshouldbestoredat+4°C,notrefrozen,andpreferablyusedthesameday.
